What sample types are supported for multiomics on nCounter?

Initial offerings are compatible with FFPE and FF tissue. Additional protocols for lysates will be available soon. The initial multiomics protocol will focus on whole slide FFPE tissues as well as fresh frozen tissue. Additional protocols are being developed for cell lysate and plasma sample types and will launch in 2026.

What will the data analysis solution look like for multiomics?

We will be introducing a new analysis software with multiomics analysis capabilities. One of the primary benefits of multiomics on nCounter is that the data for both analytes (RNA + Protein) is all the same direct digital counts. This makes it possible to combine data visualizations including heat maps, volcano plots as well as correlation tables that compare gene expression and protein expression. Additionally, already existing gene expression-only data can be combined with protein data from multiomics studies using the updated pipeline.

What protein content will be available at launch?

The initial protein panels at launch will include human panels focused on immune pathways and tumor signaling. The content for these panels was designed to complement our nCounter Oncology or Immunology panels, including our popular 360 series. Custom capabilities for proteins will also launch as an antibody labeling kit capable of labeling 5-10 antibodies. After the initial launch, Bruker will explore the development of additional panels. How are the antibodies in the protein panels validated?

nCounter protein panels include antibodies that have been validated at various stages of development and manufacturing. Antibodies acquired through a partnership with Abcam arrive with validation data that includes up to 10 different methods (Western blot, ICC, IHC, ELISA, IP, ChIP, flow cytometry, peptide array and dot blot). All antibodies included in the nCounter panels also have Abcam IHC associated images. Additionally, as part of development and manufacturing, BSB performs a series of validation testing. A comprehensive white paper outlining the commercial protein panel validation will be available later this year- stay tuned for more details!
